Hydrogels have been used widely in surgery and wound healing as adhesives thanks to their rapid polymerization and strong adhesion. The aim of this study is to synthesize a new hydrogel – Hyaluronan/Poly (vinylphosphonic acid)/Chitosan (HA/PVPA/CS) hydrogel – with characteristics that can be applied as bioadhesive for skin wound healing. The hydrogel was synthesized by chemical crosslink method; the results showed that it was stable at pH up to 5.5 and its optimized ratio in volume of HA (1% w/v): PVPA (10% w/v): CS (2% w/v) was 5:2:40. Microscopic observation presented its 3 dimensional network structure formed by random arrangement of hydrogel’s fibers. Fourier transform infrared spectroscopy (FT-IR) and Nuclear magnetic resonance (NMR) spectra indicated the presence of ester bonds, and ionic interaction in its chemical structure. Swelling and degradation results proved that the hydrogel had a fast swelling rate and a short degradation time. Besides, the hydrogel also had elastic property than can mimic skin surface to cover the wound. Additionally, the antibacterial results showed that the hydrogel performed antibacterial activity against E. coli. Lastly, it possessed positive effect in enhancing skin wound healing process in in vivo murine implantation experiments. In conclusion, with those characteristics, HA/PVPA/CS hydrogel was a potential material to apply as bioadhesive in skin wound healing.
