Cloning and expression of aprE gene encoding NATTOKINASE in Bacillus subtilis
Abstract
There is an increasing risk of thromboembolism all over the world, the
study of its therapeutic treatment is a necessity. Several studies and
treatments were published worldwide. Nattokinase which is isolated
from boiled soybean fermented food Natto of Japanese have been known
as a treatment of thromboembolia. Nattokinase were extracted and
commercialized as a functional food, but its price is still high for
Vietnamese costumers. The aim of this study is set up an over expression
of aprE gene encoding Nattokinase in Bacillus subtilis. A recombinant
plasmid pAX01- aprE is transformed into Bacillus subtilis to over-express
high content of recombinant protein Nattokinase. From 4 different
samples of Natto, Bacillus natto, which has high fibrin degradation activity, was isolated and screened. Applied the basic genetic methods, the recombinant plasmid were successfully created.
