dc.description.abstractThe most common cells of connective tissue in animal are fibroblasts which areconsidered cells that play critical role in tissue healing because of their functions: producing the extracellular matrix, stimulating re-epithelialization. Fibroblasts can be made not only in vitro but also in vivo. They are capable of making collagens, glycosaminoglycan, reticular, elastic-fiber and glycoproteins found in the extracellular matrix and cytokine (Hoyt, L. C.,2007). These proteins are a well-established system for analysis of fibroblast growth, migration and collagen metabolism in wound healing (Edward Bankowski, K. S., 1996). Furthermore, these proteins are included in umbilical cord extract.Besides, the umbilical cord extract contains a large amount of collagen elastin, hyaluronic acid (HA), proteoglycan, polysaccharide and glycosaminoglycan sulphate (GAGs) that is fixed in insoluble collagen matrix cells(Bacsich, P., et al., 1945). In the context of skin regeneration research, this investigation were performed with objective to evaluate the impactsof umbilical cord extract on fibroblast proliferation and collagen type I (one) expression. The five different concentrationsof umbilical cord extracts (0%, 2.5%, 5%, 7.5%, and 10%) were given different effects onfibroblast proliferation and collagen type I expression. MTT assay and Real time/RT PCR assay were used to evaluate the hypothesis.The results show that the higher the concentration of the umbilical cord extract, the higher rate fibroblast proliferation. On the other hands, collagen type I expression of fibroblast is stimulated the most by 7.5% concentration. This suggests that umbilical cord extract has influenceson skin regeneration.
