dc.description.abstractA successful in vitro fertilization (IVF) or intra-cytoplasmic sperm injection (ICSI) treatment is almost affected by the quality of the culture medium for development which relies to the concentration of serum added, especially bovine serum albumin (BSA). However, in Vietnam, the optimal level of BSA in mouse culture medium is not yet established. This is the reason why we conducted this study. Our study was divided into two experiments. In the first experiment, we researched on how various BSA concentrations in culture medium affect the preimplantation development stages of in vivo fertilized ICR mouse embryos. In the second experiment, we investigate the effects of different levels of BSA on the quality of blastocysts. In both experiments, embryos were cultured in modified Chatot Ziomek Bavister (mCZB) media containing 0 (control), 0.1, 1, 2, and 5 mg/ml of BSA. In the first experiment, the high blastocyst rate was observed in the 0.1 mg/ml and 1 mg/ml BSA group. However, in the second experiment, the nucleus numbers in the 0.1 mg/ml BSA group were significantly lower than the nucleus numbers of the 1 mg/ml BSA group. In conclusion, the optimal benefit to in vitro culture of 2-cell mouse embryos could be maintained when BSA was used as a protein source at 1 mg/ml.
