dc.description.abstractSolid dispersion is one of the most conventional methods to improve the solubility of poorly water-soluble drugs, however it also has some limitations. The purpose of this research is to develop a method to increase dissolution rate of poorly water-soluble drug base on the adsorption of solid dispersion (SD) on fumed silica. Solid dispersion of Ibuprofen with different ratio of Poloxamer 407 and Aerosil® were prepared by melting method and mechanical mixing. Dissolution test was conducted using conventional paddle method in imitative gastric fluid. Physicochemical characterizations like PXRD and FTIR was performed to clarify the role of polymer and Aerosil®. Also, scanning electron microscopy (SEM) images were taken to determine the morphologic change of samples. Contact angle was measured to determine the wettability of formulations. The drug release data was measured by UV-vis spectrophotometer. The result showed that dissolution rate is proportional with the amount of Poloxamer 407 but only with exact amount of Aerosil® can improve the dissolution of SDs, the optimized formulation is F5 with the ratio of 1:3:1.5 of drug, polymer and Aerosil®. Moreover, Poloxamer 407 is proved to be the suitable polymer for this research comparing with two different polymers Poloxamer 188 and PEG 6000, in terms of drug release, morphologic characteristic as well as contact angle. Overall, the adsorption of solid dispersion on Aerosil® is proved to be feasible for immediate release formulation of Ibuprofen.
