Evaluation the method for measles virus isolation on Vero/Slams cell culture
Abstract
This study aimed to Evaluate the measles virus isolation method on Vero / SLAMs cell culture to get best procedure with necessary data for laboratory quality assurance and provided technical data for isolation method of Measles virus performing in National/WHO Measles Laboratory at Pasteur Institute in Ho Chi Minh city. RT-PCR was used as an accurate method to detect measles virus by using standard kit: Supercrip III One step RT PCR system with platinum Taq DNA polymerase lifetechnologies. 30 standard positive specimens and 60 standard negative specimens were used to give out high results in the sensitivity (43%) and specificity (100%), one standard positive with measles virus to carried out experiment to determine low limit detection (10-6); using two standard positives and one negative with measles virus to proceed experiment for precision. The evaluation of the technique gives out the result and data to ensure the best procedure and conditions for measles virus isolation.
