Effects ionomycin and 6-demethylaminopurine on the parthenogenetic development of in vitro matured porcine oocyte
Abstract
The aim of this study was to survey the optimal time for treatment of ionomycin
and 6-dimethylamnopurine (6-DMAP) in parthenogenetic embryo activation. For
ionomycin testing, matured oocyte was treated with ionomycin in 3 different
duration (1, 5, 20 mins) and 6-DMAP in 4 hours , A control sample was run
alongside treatments without using ionomycin. Results in cleavage ratio was
obtained in treatments at 1, 5 20mins vs control (23.879% , 70.597 , 83.824 vs

Ratio of block ( 20.505% ,78.978% , 37.210% vs 0%) and ratio of
blastocyst ( 0% , 15.051% , 4.231% vs 0%) . For 6-DMAP testing, the best
result from ionomycin treatment was applied as standard time ( 5mins). After
matured oocyte was treated with ionomicin (5mins) , they were treated with 3
different duration ( 1, 4,10 hs), A control sample was run alongside treatments
without using 6-DMAP. The results in cleavage ratio obtained in the treatments
at 1, 4, and 10hs vs control (22.092%, 70.597%, 13.764% vs 11.970%) , block
ratio (62.179%, 78.978%, 0% vs 0%), blastocyst ratio ( 3.905%, 15,051%, 0%
and 0%). It was concluded that association of ionomycin at 5 minutes and 6-
DMAP at 4 hours was the best time duration for blastocyst development
