This study investigates antioxidant and anticancer activity of Gleditsia ausralis hemsly and two flavonoids, vitexin (GL-01) and isovitexin (GL-02), 1 sterol, β-Sitosterol-3-O- β glucopyranoside (GL-03) were purified and identified from ethyl acetate extract. Their structure were determined by NMR (1D and 2D NMR) and compared with other published data. The antioxidant potential was evaluated by 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. The anticancer activity was determined by applied RSB assay using two cancer cell line MCF-7 and HepG2. The EE had potential in anticancer activity tested with MCF-7 cell line but not with HepG2 cell line. The GL-01 showed a weak ingredient with the I% of 3.25 ± 4.00 μg/mL when screened the cytotoxicity using MCF-7 cell line and no effect on antioxidant assay. The ME had a high potential in anticancer activity with IC50 was 24.88 ± 0.51 μg/mL in HepG2 cell line. Key words: Gleditsia ausralis hemsly, Vitexin, Isovitexin, β-Sitosterol-3-O- β glucopyranoside, DPPH, RSB, MCF-7, HepG2.en_US
