Actinorhodin (ACT) showed antimicrobial activities on Mycobacterium tuberculosis and Staphylococus. Act is a blue-pigmented antibiotics produced by Streptomyces coelicolor. Actinorhodin activating gene plays a key role to synthesize blue-pigmented antibiotics. SCO5088, an actinorhodin activating gene in Streptomyces coelicolor, has been structurally sequenced but not expressed and functionally for further study. Consequently, this research was aimed at overexpression and evaluation of this actinorhodin activating gene. The objective was achieved by exploiting the E.coli BL21 (DE3) plysS host expression system. The expressed recombinant protein encoded by the gene was detected by SDS-PAGE at 46 kDa band as well as showed blue-pigmented formation in expressed E. coli. Our results reported firstly on successful overexpression of actinorhodin activating gene in E. coli.
