dc.description.abstractThe sugarcane cultivar POJ 2878 was studied to establish in vitro regeneration protocol for genetic transformation by using callus derived from immature leaves as explants. Callus initiation and growth was evaluated on MS medium containing with different concentrations of 2,4D. Callus was best induced on the MS medium containing with 2 mg/l of 2,4D. For shoot regeneration, the calli were then subcultured on MS medium added with different concentrations of BA. The best induction of shoot was observed on medium added with 1 mg/l of BA. To determine the optimum response of root formation, shoots were subcultured on MS medium supplemented with different concentrations of NAA. The result showed that root formation was highest on MS medium supplemented with 2 mg/l of NAA. Keywords: Saccharumofficinarum L.,callus, immature leaves, regeneration, shoot, rooten_US
