dc.description.abstract | The evaluation of relationship of genetic Baculovirus based on extracted DNA from insects. Its genes late expression factor 8 (LEF-8), late expression factor 9 (LEF-9), and polyhedrin/granulin (polh/gran) are highly conserved baculovirus gene. In this project, the LEF-8 segment sequences are used to identify lepidopteran-specific baculoviruses and study their diversity through molecular phylogenetic analyses. After extracting the LEF-8 segment, it were amplified by using PCR method with sets of specific primers and sequenced to estimate genetic diversity and phylogenetic relationship of Baculovirus in the Southern Vietnam and other Baculovirus groups in the world. The analysis of genetic distances showed that the Vietnamese Baculovirus and Alpha-2 Baculovirus have short genetic distance, suggesting that Baculovirus in the Southern Vietnam had close relationship to Alpha-2 Baculovirus. In addition, the phylogenetic tree also supported to above results, the Vietnamese Baculovirus were grouped in one clade with Alpha-2 Baculovirus with the bootstrap value of 93%. The Baculovirus in Southern Vietnam included 2 groups: firstly, the clustered VN1 in the group of Helicoverpa amigera with the bootstrap value of 57%; secondly, the VN2-VN8 was grouped in Helicoverpa zea with the bootstrap value of 93%. These results demonstrated that there were at least two groups of Baculovirus including Helicoverpa amigera and Helicoverpa zea which were distributed in the Southern Vietnam.
