dc.description.abstractQuality of mature oocytes has an important role in in vitro fertilization (IVF) and enhances the blastocyst formation. Thus, good mature oocytes induce the normal fertilization and also promote the developmental competence of the embryo. In order to achieve high mature oocytes quality with full cytoplasmic and nuclear maturation, dibutyryl cAMP (dbcAMP) and Follicle Stimulating Hormone (FSH) were supplemented into the in vitro maturation medium. This study investigated the effects of dbcAMP 1mM and FSH (0.01IU/ml and 0.02IU/ml, respectively) on the meiotic competence and in vitro maturation (IVM) sub-sequence in vitro fertilization (IVF) of fully grown oocytes (4-6mm). Firstly, the suitable concentration of FSH (0.01IU/ml and 0.02IU/ml) combined with dbcAMP 1mM were evaluated on the maturation rate and cumulus expansion of the fully grown oocytes. Although there was significant difference on cumulus expansion and maturation rate between basic group and supplemented groups (p<0.05) but these rates were similarly between two concentrations of FSH 0.01IU/ml (74.2% and 76.3%, respectively) and FSH 0.02IU/ml (65.9% and 74.4%, respectively) in combination with dbcAMP 1mM. Thus, the suitable combination, dbcAMP 1mM and FSH 0.01IU, were further studied on the IVM subsequence IVF. The combination of dbcAMP 1mM and FSH 0.01IU increased the normal fertilization rate significantly (39.3%) when compared with basic group (24.2%), (p<0.05). Therefore, this research is strongly claimed that fully growth oocytes cultured in maturation medium supplemented with combination of dbcAMP 1mM and FSH 0.01IU/mL possessed high maturation rate and normal fertilization.
