dc.description.abstract | Implementing in vitro fertilization (IVF) technique is essential to continue the quality of reproductiveness. IVF technique includes super ovulating females and fertilizing the oocytes with sperm which is collected from dissected epididymis of males. The purpose of this study was to investigate the effects of different concentrations of spermatozoa from young and aged male mice and pre-incubate culture of spermatozoa on the rate of in vitro fertilization and pre-implantation development of IVF embryos by using ICR mouse as a model. The first experiment was conducted to evaluate the semen quality between eight weeks old mice and one year old mice. In this experiment, the semen was analyzed through criteria: motility, morphology and concentration of spermatozoa. The result showed the correlation between semen quality derived from two aging of mouse and the ability of preimplantation development. Furthermore, this study also investigated how the sperm capacitation - the preincubate culture affected to the rate of IVF. In addition, the rate of IVF was compared through the number of oocytes fertilized by different sperm concentrations: 104,105,106 spermatozoa/ml. The findings of this study demonstrated that pre-incubate culture of spermatozoa before fertilization increases the rate of the in vitro fertilization and the optimal sperm concentration of 105/ml enhance the pre-implantation development of in vitro fertilized embryos in ICR mouse.
