Generation of arabidopsis transgenic plants over-expressing 35S:GmRR34 using agrobacterium-mediated transformation by floral dip method
Abstract
Among the abiotic stresses, drought stress is one of the most adverse
environmental factors that plants commonly have to cope with. Drought impact can
result in significant reduction in crop productivity worldwide. Recently studies
revealed that GmRR34 has an important role in plant responses to abiotic stresses,
particularly to drought. Therefore, development of transgenic plants carrying this
gene could be considered as a promising approach for improvement of plant
tolerance capacity to abiotic stress. In this project, Agrobacterium strain EHA101
carrying recombinant binary vector pBI121-GmRR34 was transferred into
Arabidopsis plants by floral dip method. The transgenic seeds from these plants
were screened on the germination medium containing kanamycin antibiotic for
selection. Transformation efficiency was 0.91% with 8 transformants out of 880
screened seeds. Two of these eight transformants were performed with PCR to
confirm the presence of GmRR34.
