dc.description.abstract | Abiotic stresses including drought, salinity, cold, and heat are known as major causes of crop failure and limitation of agriculture area. Recently, a number of TCS-encoding genes have been reported to be positive regulators in response to various abiotic stresses and suggested as promising candidate genes for developing crops with better stress tolerance. Among these genes, GmHP08 was chosen as a target for being overexpressed in transgenic Arabidopsis plants in serving for further study about its function. In this project, the floral dip method using Agrobacterium was employed for transformation. Progeny seeds were collected and then screened on germination medium containing kanamycin 30μl/ml for identification of transgenic plants. After that, to ensure presence of the transgene GmHP08, PCR was performed for putatively transformed Arabidopsis plants growing on the selective agar medium. At the end of this project, 23 GmHP08- carrying Arabidopsis plants were detected from 1918 progeny seeds, therefore the estimated transformation efficiency was 1.2%.
