Examine molecular genetic diversity of Phu Quoc ridgeback dogs by using SSR markers
Abstract
Phu Quoc ridgeback dog is one of Vietnam valuable dog breeds which carry many good characteristics such as intelligence, strength bodies, friendly with human and aslo still keeping some characteristics for hunting. Phu Quoc ridgeback dog is one of three breeds, including Thai ridgeback dog and Rhosedian ridgeback dog is classified based on the ridge hair running along their back. But the origin of them is still unclear. For a long time, Phu Quoc ridgeback dogs are facing with the genetic regression because of genetic disorders, close-blood breeding and decresing of pure dog but there is no effective solution established to protect and conserve them. The main reason comes from the lack of information about them. In this study, nucleus DNA had been extracted from 20 Phu Quoc ridgeback dogs to amplify with 6 SSR markers (ISAG, 2014) to understand allelic polymorphism of non-coding region. The amplified product was analyzed by running non-denaturing polyacrylamide and staining by silver staining method. This study would provide more information about Phu Quoc ridgeback and also examine the suitableness of SSR markers to applying in genetic test such as genetic polymorphism, especially allelic polymorphism in Phu Quoc ridgeback dog.
