Establishment of fibroblast cell lines from different tissues in the mouse
Abstract
Cell culture technologies are basic tools for most, if not all molecular biologists, and maintenance of cell for long term use. Fibroblast cell can be cultured from different tissues but they require dissimilar treatment to obtain sufficient numbers of cells. Furthermore, some kinds of cell can grow well on bacteria cellulose (BC) membrane to form wound-healing device as a scaffold. However, in Vietnam procedure to culture fibroblast cell from tail, lung and kidney of mouse as well as facilitate them integration into the cellulose scaffold are not yet investigated. Therefore, in this study we try to establish fibroblast cultures from different tissues and use as abundant sources for biomedical study. In the first experiment, we provide an easy and economical way to culture fibroblast cells from tail bone, kidney and lung of mouse. Secondly, fibroblast cells were maintained using subculture and its survival ability was confirmed by DAPI staining. Finally, develop the ability to survive and growth of fibroblast cell on BC membrane was examined. The study found that, there are many cells grown confluent from tissues and subculture is the helpful way to prolong the life-span of cell for long term studies. Also, fibroblast cell was grown well on BC membrane. They can be used for various biomedical applications.
