dc.description.abstract | Water deficit is one of the most serious disadvantages for agricultural production worldwide and is projected to be worsened under effects of unanticipated climate change. For years the scientists have been trying to understand and dissect the mechanisms of plant tolerance to water deficit using a variety of approaches; however, success has been limited. Previous studies revealed that GmNAC085 might be positively associated with the plant tolerance degree towards drought stress. Therefore, purpose of this research project is transferring the soybean GmNAC085 gene into Arabidopsis for subsequent detailed analysis of the gene function. This can hopefully generate the transgenic plants which can adapt better with drought stress and thus important for genetic engineering.
The experiments covered the transformation of construct carrying GmNAC085 gene into Arabidopsis plants and screening the transgenic. Immersion of immature Arabidopsis inflorescences in a floral dip medium containing Agrobacterium harbouring gene of interest, sucrose and surfactant was sufficient to produce transformed offspring. Selection of the transformants from non-transformants by using kanamycin antibiotic followed by the confirmation using PCR was proposed. Finally, the transgenic seedlings should be transplanted from the selective medium to soil for subsequent growing until obtaining homozygous transgenic plants.
