dc.description.abstractColorectal cancer (CRC) is the malignancy developing in the large bowel which is one of the most common malignancies and one of the leading causes of cancer death worldwide. The early detection methods or markers of colorectal cancer are necessary to improve the survival rate of CRC patients. Nowadays, new molecular tools have been developed to acquire further knowledge on tumor progression. In this study, the comparative protein expression analysis of 10 paired colorectal cancer and its corresponding non-cancerous tissue samples were performed by using the combination of iTRAQ labeling and mass spectrometry. Based on the MS/MS spectra analyzed by ProteinPilot software, 684 proteins were identified, out of which 215 were observed to be differentially expressed in at least 1 sample pair. Individual protein expression and variation have been established for each patient. Further analysis with Ingenuity Pathways Analysis (IPA) system of the obtained proteins demonstrated cytoskeletal signaling as the top-ranked functional pathway network associated with the oncogenic function. Down-regulation was observed in most of the proteins belong to this pathway such as: Caldesmon, Calponin – 1, Transgelin, Tropomyosin, etc. Besides, our research also found other interested proteins which showed significant difference in expression including Alpha-crytallin B chain (CRYAB), Calreticulin, Creatine Kinase B level, Heat shock protein beta-1 (HSPB1) and Protein disulfide isomerase (PDI). Their possible contributions to the molecular mechanism behind disease development were described. Our study supplemented the understanding about proteome of Vietnamese CRC patients and identified statistically protein expression differences among samples which can assist further studies in finding an effective biomarker for CRC diagnostics in Vietnam.
