Effect of histone deacetylase inhibitor (Trichostatin A ) on preimplantation development of clonded porcine embryos
Abstract
Successful cloning Somatic Cell Nuclear Transfer (SCNT) required epigenetic state differentiated nucleus of donor cell converted into embryonic totipotent stage. Therefore low success rate in SCNT because of failure epigenetic modification. Many studies demonstrated effects of Trichostatin A (TSA) a type of Histone Deacetylate Inhibitor (HDACi) improved the development of cloned SCNT embryos leading to investigate the effects of HDACi on cloned Bos Gaurus embryos (iSCNT). In this study, we studied the effects of different concentration of TSA (5nM, 25nM, 50nM and 150nM) for 20h after activation on preimplantation development of cloned Bos Gaurus embryos. The results indicated that treatment of cloned Bos Gaurus embryos with 25nM TSA increased the success rates of early preimplantation of cloned Bos Gaurus embryos from 8-cell stage to the 16-cell stage embryos (23%). Moreover, the immunostaining demonstrated TSA treatment significant increased acetylation signal leading to enhance transcription activity of early cloned Bos Gaurus Embryos. In summary, TSA treatment at 25nM for 20h enhance expression of histone H3K9 acetylation and improve the early development of cloned Bos Gaurus embryos.
