Primary detection of biological activities of micrococcus luteus isolated from foods
Abstract
Micrococcus luteus was known the historical interest bacterium in biotechnology with valuable properties (Smith, et al. 1999). This study was performed to develop the Micrococcus products. Primary detection of biological activities were suggested. The antimicrobial activity was done by applying difference method and checked with 95% confidence interval (95%CI). The 95% CI of diameter zone mean of paper-disc test was 8.2103 to 8.7341 in Candida albicans ATCC 10231 (P3) and
7.7537 to 8.3019 in Staphylococcus aureus ATCC 63324 (SA). The 95% CI of
annular radius zone mean of paper-disc test was 0.4009 to 0.5991 in P3 and
0.2525 to 0.4142 in SA. The 95% CI of annular radius zone mean of cylinder test was 1.8382 to 2.1618 in P3 and 1.6227 to 2.0717 in SA. The supernatant samples
of grinded Micrococcus cell had 95% CI the annular radius zone mean belonged to
range 1.9292 to 2.1264 in P3 and 3.1038 to 3.8962 in SA. The result showed the presenting of microbial agent. It had effect to affect to positive (Staphylococcus aureus ATCC 63324) and negative bacteria (Candida albicans ATCC 10231). And the antimicrobial agent was intracellular protein that only found in the supernatant
of Micrococcus cell after grinded. SDS-PAGE analysis was done to check the
possibility antimicrobial protein. The bands at 30,000Da and 50,000Da were detected in only grinded Micrococcus pellet sample. Nuclease activity detection was done on Micrococcus luteus DNA after collecting the supernatant. The digestive result was checked on agarose gel electrophoresis. The bands of Micrococcus DNA and DNA digested were detected. The results indicated the nuclease produced in
Micrococcus luteus.
