dc.description.abstract | Breast cancer is one of problem related to human health, which the world is considering nowadays. It is the most common and dangerous cancer in woman world wild. SNPs linked to the breast cancer have been identified in Genome Wide Association studies. Among those, SNP rs569550 in LSP1 gene are found to important markers for breast cancer susceptibility. The main purpose of this study is developing an optimal HRM method condition for genotyping and estimating the frequency of SNP rs569550 in 100 Vietnamese breast cancer patients. The HRM method was optimized based on the concentration of PCR reagent such as DNA, Mg2+, primer to get the optimal condition. The results were measured and evaluated through three parameters: sensitivity, specificity and stability. The optimal HRM protocol is quite efficient with high sensitivity (96%), high stability (Pvalue>0.05) and high specificity (Pvalue<0.001). Then, the Vietnamese data from 1000Genomes was used to predict the association between SNP rs569550 and breast cancer risk. Analysis of 100 breast cancer samples revealed that the frequency of major allele G was 68% and the frequency of minor allele T was 32%, thus SNP rs569550 is highly polymorphism in Vietnamese breast cancer patients. The frequency of minor T allele supports to the further association study to identified the genetic markers for early diagnosis of breast cancer for the Vietnamese population.
