Effect of histone deacetylase inhibitor (Scriptaid) on nascent mRNA expression and preimlantation development of cloned bovine embryos
Abstract
Since the first birth of Dolly, the first animal clone by somatic cell nuclear transfer (SCNT). However, the success rate of cloning animal has remained very low, and one of the most critical factors affecting success rates is epigenetic abnormalities. In this study, scriptaid (SCR) is a potential Histone Deacetylase inhibitor (HDACi) was used to examine expression of nascent mRNA and preimplantation development of cloned bovine embryos. There are three experiments were proceeded. The first experiment, the effect of different concentration of SCR (0 nM, 100 nM, 250 nM) on preimplantation development cloned bovine were examined. The results shown that at 250 μM, high rate of cloned embryos developed to the blastocyst stage (29.4%). In contract, at 100 μM, only 23.1% cloned embryos developed to the blastocyst stage. In the second experiment, the effect of duration of SCR treatment (0h, 10h, 15h) were done and the results shown that group of 10h treatment was better than group of 0h and 15h treatment for blastocyst formation (14%, 0% and 0%; respectively). In the final experiment, effect of SCR treatment on nascent mRNA expression of cloned bovine embryos were performed. The resulted shown that the level of nascent mRNA expression at the 8-cell stage of cloned bovine embryos in group of SCR treatment was higher than in group without treatment. In conclusion, this study found that treatment of SCNT bovine embryos with 250 nM SCR for 10h enhance the development cloned bovine embryos to the blastocyst stage and increase the expression of nascent mRNA at the 8-cell stage in cloned bovine embryos.
