dc.description.abstract | Trichostatin A, a potent and reversible inhibitor of Histone Deacetylation Inhibitor groups (HDACis), acts as an epigenetic modifier and results in hyperacetylation of core histone which modulates chromatin structure leading to gene transcription activity increasing.
This study was carried out to evaluate the efficiency of the concentration of histone deacetylation inhibitor (HDACi) Trichostatin A on the preimplantation development and level of nascent mRNA transcription of cloned bovine embryos. The experiment was divided into 2 parts: firstly, somatic cell nuclear transfer bovine embryos was treated with TSA to investigate the optimal concentration for improved preimplantation development. Secondly, the expression of nascent mRNA was examined by immunostaining.
Cloned bovine embryos were continuously treated with 2 different concentration 10nM and 25nm 10h starting from the activation point. The results showed that both 10nM and 25nM (17% and 18%, respectively) concentration of TSA treatment were not significantly different compared to the control group (18%) but there was an improvement on the number of cells among 10 nM group and others. Moreover, treatment with 10nM enhanced the transcription activity.
