dc.description.abstract | Major histocompatibility complex (MHC) or human leukocyte antigen (HLA) is a cell surface
molecule encoded by a large gene family. There are many types of HLAs in human. These types
are very important in transplantation and it has been known that HLA types are associated with
some diseases such as Diabetes type I, Rheumatoid arthritis, Addison disease, etc. However, there
is very little information in Vietnam about the relationship between HLA polymorphism and type
of diseases as well as statistics data on HLA frequency. This study aimed to investigate the HLA
– II polymorphisms among healthy donors and end – stage renal disease patients tested at Cho Ray
Blood Transfusion Center in Cho Ray Hospital to provide some preliminary evaluation whether
there is an association between HLA – II polymorphism in Vietnam population and the
susceptibility to diseases as well as comparing HLA frequencies data collected in Cho Ray hospital
with that in different population around the world. Collected blood from end-stage renal diseases
and donors will be used to extract DNA. Then PCR are used to amplify DNA. After that, the PCR
products were used to perform denaturation, neutralization and hybridizing. Finally, HLA type was
determined based on the calculation and compare the beads with positive and negative values of
the samples with the available in LABType-SSO form. This study was among the first ones to
provide information on HLA-II of Vietnamese people. The results showed evidence on the
relationship between HLA – DRB1*12 and Rheumatoid arthritis, HLA – DRB1*03 and HLA –
DRDQ*03 linked to Diabetes type I and HLA – DQB1*03 had associate with Allergic rhinitis.
Additionally, HLA DRB1 *12 (30%), HLA DQA1*01 (32%), HLA DQB1*03 (60%) had the
highest appearances frequency in ESRD patients. Similar results had been pointed out in healthy
donors for highest appearance frequency HLA DRB1 *12 (28%), HLA DQA1*01 (33%), HLA
DQB1*03 (51%). Our study suggested that HLA type II test can provide early disease dianogsis,
hence, increasing the potetial of disease prevention.
