Synergistic effects of trichostatin A and scriptaid in one step treatment on transcriptional activity and development of cloned bovine embryos
Abstract
Recently, the application of histone deacetylase inhibitors has made it possible to
improve the cloning efficiency in many species. This study investigates the
synergistic effects of Trichostatin A (TSA) and Scriptaid (SCR), the combination
of two most popular histone deacetylase inhibitors (HDACi), in one step
treatment on preimplantation developmental competence and transcriptional
activity of cloned bovine embryos. Single treatments with 250nM SCR; 10nM
TSA and combined treatments with 125nM SCR and 5nM TSA for 10h; 52h after
activation were compared to evaluate their effects on the development rate of
SCNT embryos. As the results, the blastocyst formation of the combined treated
group for 10h post-activation was significantly increased compared to combined
treated group for 52h post-activation and single treated group with TSA,
however, was not higher than single treated group with SCR. Then, to observe
the transcriptional activity of cloned embryos, immunofluorescence staining was
performed, finalized that the level of nascent mRNA was highly expressed in 8-
cell stage embryos of the combined treatment. In summary, this project would
contribute to uncover mechanisms underlying the synergistic effects of TSA and
SCR on the improvement of bovine SCNT embryos
