Cloning An Aa13 Polysaccharide Monooxygenase Gene Into The Pex2b Vector
Abstract
Polysaccharide monooxygenases (PMOs) cleave glycoside linkages in polysaccharides,
which have great potential application in the manufacturing of biofuel from
polysaccharide biomasses. Our objective is to insert the PMO genes into the plasmid
pEX2B to express PMOs in filamentous fungus for improving the application PMOs into
the industry. In this study, we successfully designed primers and amplified PMOs
AA13 gene from Myceliophthora thermophila genome and then PMOs AA13 gene was
cloned into pEX2B vector for transformation into DH5α competent cell.
