Syzygium cumini (Linn.) leaves contain a high level of flavonoid and phenolic compounds which display antioxidant ability. The study is aimed to evaluate the potent antioxidant activity of S. cumini leaf extract in in-vitro and in-vivo conditions. The effect of consumption of S. cumini leaf extract was justified using in vitro DPPH, ABTS, reducing power assays and on H2O2-induced oxidative stress Drosophila melanogaster flies with MDA and GSH as biomarkers. D. melanogaster was exposed to a concentration of 0.25 and 0.5 mg/mL of the leaf extract. The in vitro results showed that the leaf extract possesses antioxidant characteristics despite smaller capabilities than vitamin C. For the in vivo, it was suggested under non-stress conditions, the leaf extract can increase longevity for males, but had a negative effect on the life cycle of the females. Under oxidative stress conditions, the same pattern could be observed. For most cases, 0.5 mg/mL had the tendency to express the best, the higher the concentrations, the better the ability to fight radicals. However, sometimes, 0.25 mg/mL was a better performer depending on the tests and the gender. The obtained results pointed to the potential of S. cumini leaf extract used for prevention and possible treatment caused by oxidative stress, but it still needs further investigation.en_US
