dc.description.abstractThe ability of endophytic fungi to produce valuable bioactive compounds when surviving in medicinal herbs. Finding out the biological sources is essential for pharmaceutical field. Endophytic fungus was isolated from Angelica sinensis originated in Kon Tum province (Vietnam). The isolation was based on the morphology of fungi. Identification was performed by sequencing 18S rRNA that was compared with known genes using Blast search in the combination of phylogenic analysis by using ClustalW and PhyML in GenomeNet. The supernatant of isolated fungi was extracted and used to test antimicrobial activitiy, which include antibacterial and antifungal activities. Antibacterial activitiy on Staphylococcus aureus ATCC 6538, Pseudomonas aeruginosa ATCC 27853, Micrococcus luteus ATCC 9341 and Klebsiella pneumoniae ATCC 10031 were carried out by well diffusion method. In terms of antifungal activities on Aspergillus sp., Fusarium sp. were carried out by micro dilution method. As a result, Acremonium sp. F01 and Aspergillus sp. F02 were identified. All two isolated endophytic fungi extracts showed biological activities. With antibacterial test, the extract of isolated fungi strain F01 in 150 μg/mL and 200 μg/mL concentration had the strongest inhibitory effect on K. pneumoniae (13.37 ± 0.61 and 16.53 ± 0.72 mm) than S. aureus (10.33 ± 0.31 and 12.10 ± 0.56 mm), M. luteus (8.13 ± 0.40 and 9.93 ± 0.50 mm) and P. aeruginosa (7.24 ± 0.31 and 9.70 ± 0.40mm). Especially, when the effect against K. pneumoniae was compared to Ciprofloxacin 5 μg/mL, F01 with the concentration of 200 μg/mL gave inhibition zone that was higher than positive control (16.53 ± 0.72 and 15.88 ± 0.61, respectively). With antifungal test, the extract of isolated fungi strains F01 and F02 indicates high antifungal activity against Fusarium sp. at 256 μg/mL concentration of MIC. With Aspergillus sp., the bioactive of F01 and F02 strain illustrated antifungal activity with MIC values of 16 μg/mL and 32 μg/mL, respectively. In conclusion, the study suggested fungus isolated from Angelica sinensis could show and give many biological compounds that will be a potential source for pharmaceutical field.en_US
