Evaluation Of The Yield Of Total Soluble Solids Content From Đảng Sâm (Codonopsis Pilosula Nannf.) By Using The Combination Of Enzyme And Ultrasound Treatment On Conventional Extraction Method
Abstract
The objective of this study was to evaluate the effectiveness of extractions from
Codonopssis pilosula Nannf under the treatment of Viscozyme® L and ultrasound asisted on the conventional method was investigated to evaluate their effects on the
total soluble solids conent (TSSC). The methods of this study included testing the
wide range of duration of extraction (0-6hrs), enzyme concentration (0-2.5%),
duration of enzyme incubation (12-36hrs), amplitude of ultrasound (0-60%), and
ultrasonic duration (10-40mins). The highest total soluble solids content was 8.17 ±
0.37oBrix under 5hrs for extraction duration, 1.5% for enzyme concentration, 24hrs
for enzyme incubation duration, 50% for ultrasound amplitude, and 30mins for
ultrasound duration. Furthermore, the total soluble solids content in combined
methods (enzyme and ultrasound) were significantly higher than those in single method extraction at p<0.05, which pointed out the dramatically effect of combining
enzyme and ultrasound extraction. Other parameters, including TSC, TPC, TFC, TSP,
TSS, and RD were investigated through biochemistry tests on the sample with the
highest solute content. TSC was 104.71 ± 5.20 (mg/g), TPC was 11.77 ± 0.52
(mg/g), TFC was 5.28 ± 0.14 (mg/g), total TSP was 39.34 ± 1.61 (mg/g), TSS was
197.05 ± 9.84 (mg/g), and RS was 171.61 ± 8.57 (mg/g). The contribution of this
project was set an insight to the positive effect of combination of extraction method.
In addition, it also suggests a possible improvement in the current rudimental industry
of Đảng Sâm (C. pilosula)
