dc.description.abstract | The objective of this study was to evaluate the conditions to produce cellulase, using Aspergillus fumigatus culture, and applied the produced enzyme in enhancing total saponin extraction from Đảng sâm (Codonopsis pilosula Nannf.). The methods of this study included microbiology culture of fungi (e.g revival microorganisms, enumeration, etc.), extraction methods (e.g. solvent and enzyme-assisted extraction), biochemical analysis (e.g. refractometer, colorimetric spectrophotometer). The crude enzyme produced by A. fumigatus Viscozyme L was used before solvent extraction to break down the cell wall of sample, in order to improve efficiency. The most favorable conditions were obtained by incubation at 55oC for 6 with 80% water content. The results showed that the enzyme produced by A. fumigatus culture was proved to have similar effects in cellulytic activity compared with Viscozyme L. In both cases, pretreatment ginseng samples with enzymes for 2 hours resulted in 1.8 times enhancing in the amount of total saponin, and 3 times in the amount of total soluble solid contents than the control sample. | en_US |
