dc.description.abstractIn this study, project mainly focused on investigating the development of cloned bovine embryos in preimplantation as well as the expression of histone acetylation during the first cell cycle after changing the location of somatic cell nucleus which was injected into enucleated mature bovine oocytes (Somatic Cell Nuclear Transfer, SCNT). In each group, the control groups were used for transferring cell nuclear in the enucleated area (the location of metaphase II), while the experiment groups were designed for injecting in different locations, such as opposite (1800 degree) and square (900 degree) angle sites to the location of metaphase II. The research was designed for 2 experiments, which are to evaluate the preimplantation development of cloned bovine embryos in different positions of somatic cell nucleus injection into enucleated oocytes; and analyze histone acetylation expressed in the first cell cycle (10 hours of post-activation) of those location. In the first experiment, the preimplantation development was only approximately 23% in blastocyst formation. Besides, there were no significant differences from 2 cell stage to blastocyst stage in development and survival rate (p>0,05) between each group. In the second experiment, the location of fibroblast cell injection at 90o direction and 180o direction did not impact the expression of histone H3 lysine 9 acetylation (acH3K9) based on the intensity in each group (p>0,05). Moreover, all of those still form the pronuclei in the first cell cycle, leading to the similarity in all groups of the experiment. In conclusion, changing the location injection during SCNT does not have any impact on the development of the SCNT bovine embryo during preimplantation as well as the H3K9Ace modificationen_US
