dc.description.abstract | Embryonic stem (ES) cells possess the remarkable capacity to undergo differentiation into
a wide range of body cell types. It is proven that single mouse blastomeres can derive ES
cells using a PGD-like technique. Studies have demonstrated how crucial culture
conditions are to the effective derivation of embryonic stem cells from single blastomeres.
In this study, I focus on detecting the effect of different culture media on ES derived from
ICR mouse single blastomeres, specifically modified Chatot Ziomek Bavister (mCZB)
medium and stem cell (SC) medium. The 8-cell ICR mouse embryos were biopsied using
the combination of XYClone Laser and micromanipulator system. Following, the isolated
blastomeres were cultured in mCZB-SC medium and SC-SC medium. The using of a basic
SC culture medium resulted in 30,4% of ES cluster formation in the SC-SC medium
experiment, and 28,57% in the mCZB-SC medium. The studies show that mCZB medium
supported preimplantation development of single blastomeres, but pre-culture did not
increase ES cluster formation rate in SC medium. Continuous culture post-biopsy
improves ES cluster formation. | en_US |
