dc.description.abstract | Nàng Thơm Chợ Đào (Nang Thom Cho Dao) is one of the Vietnamese traditional rice varieties widely cultivated in Cuu Long river Delta; it is the most aromatic rice variety among them. People know it as the fragrance and plasticity in cooking quality and high consuming market of the variety in Vietnam have been always noticed. In breeding, in addition to the traditional methods, the biotechnological methods have been applied to improve some traits such as highly susceptible to lepidopteran insects in monsoon season, plant height,… In this trend, first, a complete system including callus induction and plant regeneration phases need to be established in the in vitro condition.
De-husked seeds were rinsed with distilled water for 2 - 3 times, then sterilized by the ethanol solution 70% (w/w) for 5 minutes and by the Javel solution 50% (w/w) for 30 minutes. After disinfection, they were cultured on the MS (Murashige and Skoog, 1962) basal medium containing 2,4-D (1, 2, 3, 4 mg/l) for callus induction (kept in the dark condition, temperature 25-28oC). After 1.5 months of culture, the result showed that the highest percentage of callus induction was observed in the medium supplemented with 2 mg/L of 2,4-D. The formation of globular pro-embryos of embryogenic callus was obtained, the non-embryogenic callus also appeared along with embryogenic callus during subsequent subculture. Embryogenic callus could be subcultured/maintained on the agar medium and in the liquid medium for some months.
For plant regeneration, callus (1 months after culture, about 5 mm in size) was cultured on the MS basal medium containing different concentrations of NAA and BA (kept in light condition - about 3000 lux, temperature 25-28oC). After 1.5 months of culture, the result showed that the best response of callus for regeneration of shoot/plant was observed on the medium supplemented with 1 mg/l of NAA and 3 mg/L of BA.
The regenerated plants were rooted in the MS medium without plant growth regulator prior to growing in the greenhouse.
