Establishment Of An Optimization Culture Medium For In-Vitro Development Of Mouse Embryos.
Abstract
were a lot of experiments focused on the effect of medium in the development
of in-vitro fertilized embryos. However very a little studies on the effect of culture
media on the pre-implantation development of in vivo fertilization. Therefore, the aim
of this study was to test how difference of two media (CZB and KSOM) on the
development of in vivo fertilized 1-cell embryos during pre-implantation
development. Female mice (8 to 10 weeks old) were super-ovulated using PMSG/hCG
and mated with a proven fertile male of the same strain. Embryos were collected and
cultured in vitro around 26 h after hCG injection. Embryo development was evaluated
at both 96 h and 120 h after hCG injection. The result showed that the CZB medium
was better than the KSOM medium on the development of in vivo fertilized ICR
mouse zygote to the expanded blastocyst and hatching blastocyst stage. There
should be a concern to use CZB medium for in-vitro culture of mouse embryos.
