Study on the chemical constituents and bioactivities of angelica sinensis
Abstract
This study aims at isolation of chemical constituents from Angelica sinensis root and evaluation of their bioactivities. Three isolated compounds were determined as β-sitosterol (375.6 mg), bergapten (50 mg) and β-sitosterol-3-o-β-D-glucopyranoside (184.4 mg). DPPH assay was performed to evaluate antioxidant activity. Ethyl acetate extract and the fractions of 100% methanol (M100) exhibited the highest antioxidant activity among all extracts and fractions, IC50 were 46.20±0.09 μg/mL and 31.57±4.89 μg/mL, respectively). ANS01 compound exhibited antioxidant activity (IC50 = 47.71 ± 1.40 μg/mL). Sulforhodamine B colorimetric assay (SRB assay) was used to evaluate inhibitory activity against MCF-7 and HeLa cells growth. The methanol extract exhibited higher cytotoxicity against HeLa cell line growth at screening concentration (percentage inhibition of 19.77±5.83 % at 100 μg/mL) while n-hexane extract and ethyl acetate extract exhibited higher inhibitory activity against MCF-7 cell growth (percentage inhibition of 15.71±6.41 % and 14.94±2.00 % at 100 μg/mL, respectively). ANS01 compound had potent in anti-breast cancer activity by exhibited inhibitory activity against MCF-7 cell (percentage inhibition of 29.32±2.29 (%) at 50 μg/mL).
